BPC-157 is a synthetic 15-residue peptide, CAS 137525-51-0, formula C62H98N16O22, molecular weight 1419.53, sequence H-Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val-OH. TB-500 is the acetylated actin-binding segment of thymosin beta-4, residues 17 to 23, CAS 885340-08-9, formula C38H68N10O14, molecular weight 889.02, sequence Ac-Leu-Lys-Lys-Thr-Glu-Thr-Gln-OH.44.
In published work the three model different steps of one broad process. GHK-Cu appears in fibroblast and collagen-expression studies and in copper coordination chemistry, where the histidine imidazole and terminal amine form the site that gives the solid its blue color. BPC-157 is examined in rodent gastrointestinal and tendon-injury models. TB-500 is studied for actin sequestration and cell migration in culture.
What is not settled is whether the fixed ratio behaves differently from the same peptides assayed individually. No controlled comparison of the co-lyophilized blend against its separate constituents has been published, and none of the three has a fully mapped mechanism. The preparation is best regarded as three distinct analytes sharing a container.
Reconstitution & handling
The cake dissolves in bacteriostatic or sterile water added slowly down the vial wall, since all three components are freely water soluble. Total concentration is the labeled 70 mg divided by the diluent volume, with component concentrations following the 50:10:10 ratio. Swirl gently rather than shaking, because the long TB-500 chain foams. A pale blue tint is the copper complex and is expected.
Storage & stability
Sealed vials are held at -20 degrees Celsius, protected from light and from atmospheric moisture, since the cake is hygroscopic and a softened cake signals seal failure. Reconstituted material is kept at 2 to 8 degrees Celsius, divided into single-use aliquots, and protected from repeated freeze-thaw cycles. Two component notes apply. The copper center is redox active and photosensitive, so amber glass suits it, and the single methionine in TB-500 is oxidation prone.
How it's tested
Reverse-phase HPLC with detection at 214 and 220 nanometres resolves the blend as three peaks whose relative areas are checked against the declared 50:10:10 mass ratio rather than reported as one purity figure. Mass spectrometry confirms each component against its theoretical mass, including the copper isotope pattern. Net peptide content is measured so the 70 mg label reflects peptide rather than counter-ion and water.
